Repeatedly Checking a Coating Trial: Set the Review Rules in Advance

Set interim review and stopping rules before a coating trial so repeated checks do not turn a temporary favourable result into an overstated conclusion.

Updated · NANOLAB Knowledge

Inspecting a trial regularly is useful for safety and quality. Repeatedly running an ordinary significance test and stopping at the first favourable result is a different activity and can distort the stated false-positive risk.

Separate monitoring from inference

Define which checks look for damage, process problems or missing data, and which checks can trigger a claim of product superiority. Safety or quality stopping should remain possible even when the statistical comparison is incomplete.

Write the review plan beforehand

  • Primary outcome and meaningful difference.
  • Planned sample size or follow-up duration.
  • Timing of interim analyses.
  • Rules for stopping, continuing or adding data.
  • Method for handling repeated analyses and repeated specimens.

Use an appropriate analysis

A fixed final analysis and a properly designed sequential analysis are different approaches. Sequential methods must fit the data structure, including repeated measurements on the same panel. Obtain statistical support when the result will underpin an important performance claim.

Report deviations transparently

If the study is extended because the first result was inconclusive, record that decision and its effect on interpretation. Do not present a selectively chosen visit as the only analysis ever performed. For example, a coating leading at week four but not week eight should have both observations retained. The review plan keeps an exploratory signal useful without converting it into unsupported certainty.
